This week, nearly 370,000 bottles of duloxetine, a commonly prescribed antidepressant, were recalled after testing identified levels of N-nitroso-duloxetine above FDA’s recommended limits. The recall follows several years of similar actions involving products such as valsartan, metformin, ranitidine, varenicline, and now multiple rounds of duloxetine recalls.
For many companies, nitrosamines are viewed as a regulatory hurdle. In reality, they represent something much bigger.
They are a reminder that product quality, process understanding, analytical control, packaging, storage conditions, and supply chain management are all interconnected. A seemingly minor change in raw materials, manufacturing conditions, excipients, or storage environments can create conditions where nitrosamines form or increase over time.
The challenge is that nitrosamine risk does not disappear once a product reaches the market. It must be evaluated throughout development, technology transfer, validation, commercial manufacturing, and lifecycle management.
At Syner-G, we help clients move beyond simply reacting to regulatory requests. Our teams work with sponsors to:
- Perform comprehensive nitrosamine risk assessments for drug substances and drug products
- Evaluate synthetic routes, raw materials, reagents, solvents, and packaging components for potential nitrosamine formation pathways
- Develop scientifically justified control strategies
- Support analytical method development and validation
- Prepare regulatory responses and health authority submissions
- Build long-term mitigation plans that stand up to regulatory scrutiny
The companies that successfully navigate nitrosamine challenges are rarely the ones scrambling after a recall. They are the organizations that identify risks early, understand their processes deeply, and build mitigation strategies before regulators ask the questions.
As today’s recall demonstrates, nitrosamines remain an active area of regulatory focus. The best time to address the risk is long before it becomes a compliance issue.
